Although the 5-year survival rate has been rising over the past few decades, it is still low at about 65%. The current standard of care for colorectal cancer (CRC) involves chemotherapies developed more than 50 years ago. MicroRNAs mediate host-microbiome interactions in colorectal cancer progression As an outcome, it will fundamentally advance our knowledge of how anti-tumor immunity was generated and regulated in CRC, and provide novel targets for CRC immunotherapies.ģ. We are testing the hypothesize that ACKR4 positively affects immune regulation in CRC and confers antitumor immunity by modulating CCR7/CCL19/CCL21 chemotaxis. The results from this pilot study will establish the clinical significance of ACKR4 expression levels to immune response in colorectal cancer. This receptor, is involved in the internalization and degradation of multiple chemokines, such as chemokine (C-C motif) ligand 19 (CCL19) and CCL21, thereby modulating the CC-chemokine receptor 7 (CCR7)/CCL19/CCL21 chemotaxis and its downstream immune responses. One of the under examined features of CRC is the regulation of anti-tumor immunity by atypical chemokine receptor 4 (ACKR4) expression. Several chemokines and cytokines are implicated in CRC immune response. Atypical Chemokine Receptor 4 (ACKR4) in anti-tumor immunity in colorectal cancer Results from this study will fundamentally advance our knowledge of how cancer cells modulate and suppress the immune response, provide novel targets, and form the basis for a new anti-cancer therapeutic strategy.Ģ. We are testing the following hypothesis that extracellular vesicles (EVs) containing immunosuppressive microRNAs secreted by CRC cells suppress host T cell activity locally and in tumor draining lymph nodes, resulting in a deficient immune response that allows for increased tumor progression and growth. T cells isolated from non-immunogenic, microsatellite stable (MSS) CRC have lower levels of the CD28 protein, which provides a co-stimulatory signal required for T cell activation, trafficking, proliferation, differentiation and cytotoxic activity. they lack a significant number of infiltrating T cells, and are typically unresponsive to the immune-checkpoint inhibitor based therapies that have dramatically changed the way we treat many cancer patients. The majority (~85%) of CRC tumors are non-immunogenic, i.e. Tumor derived extracellular vesicles and anti-tumor immunity in colorectal cancer.Ĭolorectal cancer (CRC) remains the third most common cause of cancer-related deaths in the U.S. Gene expression profiles - developing novel diagnostic markers and identification of therapeutic targets in sarcomas and other cancers.ġ. This would be useful in a language learning context where you might want to add, say, the Spanish word for “tree” near a tree visible in the video. Labels add a simple text box that are best used for… labeling things.I was able to upload a custom 360 video without trouble, and immediately start adding annotated tags. The creation part couldn’t have been simpler (and in its pursuit of simplicity also feels a bit light on features). Duke is piloting the technology, so I took the opportunity to test both the creation and publishing of 360 video through Thinglink. Thinglink, as described previously, is a web platform that allows the user to add interactive pop-up graphics onto photos and videos, in 2D or in 360. However, the tool Thinglink now offers an intuitive way not only to augment media with interactive annotations, but to link that various media to each other. Neither working with an external vendor, nor developing a in-house solution seemed worthwhile for our needs. The challenge has always been how to add that interactivity. Particularly when viewed through a headset, the immersive nature of 360 video lends itself well to exploration and curiosity. Watching a 15-second old version of myself from within a VR headset is probably the closest thing I’ve ever had to out of body experience.Īs long as I’ve been working with 360 video, one element has always been out of reach: interactivity. I could alternatively input the URL manually into the browser, but that process is a bit tedious when wearing using the headset. Within the headset, I opened the YouTube app, searched for my channel, and accessed the stream from my videos there. To access the stream from in the Oculus Go, like most things in a VR headset, is straightforward if not exactly seamless. From real life to the live feed, I estimated about a 10-15 second lag. After a few moments, the feed came through, I toggled on the 360 video option, and I could then Go Live from YouTube to take the stream public. I then hit the “Live” button to send the signal to YouTube. I ran the video feed at 4k, 30FPS, 15Mbps bit rate. Back in the camera control app, I made sure it was in Custom Rtmp server, and plugged in the stream URL and key from YouTube. YouTube provided me a URL and key code to plug into my camera control app. I named it, and made sure the stream was unlisted so that only I knew where to access it. YouTube makes this really easy – there’s an icon right on the homepage that allows you to “Go Live.” From here, I set up the stream. With the camera control app on the laptop connected to the camera, I then went over to YouTube to set up the livestream host. I could also use the camera control app on an iPad or other mobile device, but using a laptop to setup the livestream was much easier since I could access both the camera application and the livestream host on the same device at the same time. I could’ve theoretically streamed just over WiFi without plugging into the Ethernet, but I found the connection wasn’t strong enough when I later actually went to livestream. I plugged that IP address into the camera control app on my laptop which was on the same local wifi network and got connected to the camera. I then connected the camera via cable to an ethernet port, which generate a new IP address for the camera. After connecting the camera to my local wifi, I updated the WiFi settings on the camera to be in Access Point (AP) mode. You can follow along this process with this video capture of setting up the stream. Though there’s a few ways you could set up the livestream, I found the following to be the most reliable. In particular, I was curious about viewing the livestream from within our VR headset, the Oculus Go. While we’ve worked with the Insta 360 Pro fairly extensively in the past, we hadn’t yet tested it’s capability for livestreaming. Many people will see a greater loss in inches around the body initially. Additionally, after weighing yourself, you should take your measurements. Every 3 weeks, you should weigh yourself in the morning before eating or drinking to track a consistent weight loss. In addition to keeping track of the food and containers you eat each day, it’s also so important to keep track of your weight loss and inches lost each round you complete. Bonus 21 Day Fix Printables and Worksheets Again, you simply print as many as you need for the full 21 day program (or however many rounds of the program you plan on doing), or wipe and re-use the same sheet if it is laminated and you use a dry erase marker with it.įor higher brackets like 1800-2099 calories and up, you can check out my shop on Etsy for the full set of tally sheets. I have the same options available for the next up calorie bracket. Visit my Portion Control Printables Shop on Etsy! 1500-1799 Calorie Bracket Printable You can print these and put them in your work agenda/planner or purse and tick them as you go throughout the day. The other option is the tally sheet that offers space to track 6 days worth of the diet. When I’m done the three days, I erase the laminated printable and start again! □ I then laminate it and use a dry-erase marker to write each day. Personally, I like to save on ink and paper and print off just one or two copies of the sheet as seen below. You can either print out a bunch of these 21 day fix tally sheets and have them in a binder to keep track of each and every day. This also allows you to “copy” meal and snack ideas on other days and remember foods that you are eating or meals that are quick to make. Some people just like to tick off the containers as they go through the day whereas others prefer to have space to write down exactly what they have for breakfast, lunch, dinner and snacks to make sure they are logging everything on their worksheet.įor those who like to log down your daily meals and snacks, I’ve created a tally sheet with space beside each meal to write what you had. I’ve designed various worksheets to use with this calorie range depending on how you prefer to proceed throughout the program. 21 Day Fix Worksheets for the 1200-1499 Calorie Bracket Once you have achieved your weight goal and are on “maintenance” mode, you will progress to the 1800-2099 calorie bracket or slightly higher depending on your activity levels. Most women who are trying to lose weight are using the 1200-1499 or 1500-1799 calorie bracket. The worksheets are designed to help you keep track of how many containers you are consuming each day depending on which calorie bracket you have chosen to follow for your diet. Looking for 21 day fix printable tally sheets to help you keep track of your progress throughout the program? You’ve come to the right place! We’ve designed a whole little shop full of affordable tally sheets for the 21 day fix Beachbody program or any other workout program using portion control containers. Each of the two factions fighting will also be assigned a commander, and the commander is in charge of not only the team and squads, but tank deployments, air strikes, and orders, however these orders cost resources. In the Offensive mode, a defending team is in control of all sectors at the beginning of the match, and the objective for the opposing side is then to capture all of them before the timer runs out. In the Warfare mode, the game is won by either controlling all sectors at a given point in time, or by controlling a majority of them when the timer runs out, essentially functioning as capture the flag. In both modes, the map is divided into sectors that each team seeks to capture and control. Īs of October 2022, two game modes exist in the game: Warfare and Offensive. Each consists of multiple smaller rifle squads of six soldiers, armoured squads of three soldiers, or recon squads of two soldiers. 50 combined arms battles between two teams of Germans and Americans, and, more recently since the full release of the game, Soviets. It was released for PlayStation 5 and Xbox Series X/S on 5 October 2021. It was initially released for Microsoft Windows as an early access title on 6 June 2019, with full release in July 2021. The game was announced via a successful Kickstarter campaign in 2017, where it raised US$220,000. Players fight in iconic battles of the Western and Eastern Fronts of World War II at the platoon level. Hell Let Loose is a multiplayer tactical first-person shooter video game developed by Australian studio Black Matter and published by Team17. Homemade India ink should only ever be used for general art, not tattoo work as the risks skyrocket due to the often unhygienic ingredients that are used in its creation so always avoid using any inks that are homemade for tattoos.Įven when using a more reputable India ink for your tattoos though, you will almost always see a dramatic improvement in the quality of your tattoo if you opt to use an actual tattoo ink instead of India ink. In our opinion, the Winsor & Newton Liquid Indian Ink is one of the better options and it is commonly used for general tattoos as well as stick and poke tattoos too.Īs we touched on earlier in the article, we know that more and more people are starting to make their own homemade India ink as it is quick, easy, and cheap. If you are set on having your India ink tattoo the there are a number of different brands that can all work well with the process and deliver decent results. If you are wanting a professional-looking tattoo to last you for a long period of time then a real tattoo ink will be a better option. This compounded with the relatively high chances of reactions and ink rejections with India ink should be enough to push most people away from it.ĭue to the pigmentation in India ink being based around soot or charcoal ash, it tends to be less black than modern tattoo ink too making it a pain for a number of tattoo styles too. In addition to that, a big risk with people who do stick and poke tattoos is that their equipment may not actually be sterile increasing the risk of problems even further. The chance of any negative side effects are drastically reduces when using an ink designed for use with tattoos and the chance of ink rejection for the majority of people is minimal. In this day and age, actual tattoo ink is often cheaper than the better India ink formulas making it much more cost-effective to just get real tattoo ink that has been designed for use with tattooing. That said though, even if you do use a none toxic India ink formula for your tattoo, there is still a relatively high risk of different types of reactions as well as ink rejection so in our opinion, it simply is not worth it. As we touched on earlier, there are a number of none toxic India ink products on the market that do tend to be fine for stick and poke style tattoos but some India ink formulas may have higher levels of toxins so always double-check the toxicity level of your ink prior to use. Is India Ink Safe For Tattoos?Īlthough India ink is technically safe for use with tattoos, there are a number of conditions that do apply that will make it increasingly risky for both potential health issues as well as tattoo quality. We know that a number of people do make their own homemade Indian ink and we would never recommend that you use homemade Indian ink for your tattoo work. In our opinion though, regular tattoo ink is almost always going to be better than using Indian ink and it is often around the same price as regular tattoo ink too. One of the more common options is Winsor & Newton Liquid Indian Ink and we know that it has been used heavily for tattoo work over the years. There is an absolute ton of India ink on the market these days with a number of them not being toxic and having been used for stick and poke tattoos for years. Our hope is that we will be able to help our readers make a more informed choice for their tattoos that not only have a lower health risk but also usually result in better looking tattoos. Although India ink has been used for decades for some types of tattoo work, mainly stick and poke with minimal issues, we still see a number of people reaching out to ask if India ink is safe for tattoos.ĭue to seeing so many people reaching out and asking for advice on using India ink for tattoo work, we have decided to publish this article going over our thoughts on using India ink for tattoos, if it is safe, as well as a number of other common questions that we see about India ink for tattoo work. Our content may have affiliate links that can result in commissions for qualifying purchases, full details in our privacy policy.Īlthough Indian ink has been used for centuries for various types of artwork, the switch over of tattoo moving from being part of certain sub-cultures to being mainstream has lead to more and more people asking about using India ink for tattoo work. Times Square is the most Instagrammed landmark in America, according to an analysis by the photo printing company Printique. Tom Harris, the president of the Times Square Alliance, thanked the mayor and then said: “You’re in Times Square more than most of our office workers, so our office workers need to step up and show up.” “We would have to raise taxes or cut services, and that’s exactly the trap that we want to make sure we don’t fall into.”Īt a news conference in Times Square this month, Mayor Eric Adams declared in a speech that “the comeback of America starts here in this square.” “Imagine if just a piece of that disappeared, how we would have to fill that gap,” said Seth Pinsky, who was an economic development adviser to former Mayor Michael Bloomberg’s administration. The pandemic wiped out more than $28 billion in value from the city’s office buildings, according to a report last year from the New York State Comptroller’s office, a potential threat to the city’s tax base and fiscal health. Many of New York City’s political and business leaders are desperate for office workers to come back. In 2016, Times Square’s economy was the same size as the city of Nashville’s. Times Square is vital to New York City’s recovery, given its concentration of office buildings, tourist attractions and hotel rooms around the city’s busiest subway station. Hotels depend on nearby office buildings to bring business travelers in for meetings, helping to fill up rooms on weekdays. Many businesses rely on commuters to spend money around the office on coffee, lunch, dry cleaning and happy hour. The surrounding blocks are home to more than two dozen office buildings. This is bad news for Times Square, where 20 percent of storefronts are still closed. Four months earlier, Michelle Go, a Deloitte employee, was pushed to her death on the subway tracks at the Times Square station. Daniel Enriquez, a Goldman Sachs employee, was fatally shot on a subway last Sunday on his way to brunch. Lately, the conversation around returning to the office has centered on public safety following a string of violent crimes on the subway. Only 8 percent were back five days a week. As of late April, 38 percent of Manhattan’s office workers were at their desks on a typical weekday, according to a survey released this month by the business advocacy group Partnership for New York City. On some days this month, there was even more foot traffic than on the same day in 2019.īut even as restaurants, Broadway shows and concerts are feeling crowded again, the office is not. More than 300,000 people are regularly walking through the neighborhood each day, about 20 percent below prepandemic levels, according to the Times Square Alliance, which represents the area’s businesses. The more crowded Times Square becomes with visitors, the more off-putting it is for the white-collar office workers who now have the choice to work from home. The problem is that’s only half the equation. Images of the eerily empty square ricocheted around the world and became a symbol of the city’s devastation as an epicenter of the pandemic.Įarly in the pandemic, an existential question facing New York City was what would still attract people to neighborhoods like Times Square.Īs it turned out, the Palace Theater would symbolize a key piece of the answer: People come to New York to have fun. In March 2020, the entire ecosystem collapsed. But that was Times Square as it was intended to be - a destination for both work and play. The revitalization of Times Square was almost too successful at attracting people, turning the sidewalks into a live-action video game where lawyers and accountants were forced to push past selfie sticks and costumed Elmos to get inside their offices five days a week. The theater where Judy Garland and Liza Minnelli once performed was now showing “SpongeBob SquarePants,” the musical. A Doubletree Hotel was built on top of the Palace Theater, heralding a booming era for tourism in the city. In the 1990s, an effort to clean up the seedy image of Times Square brought new office buildings to the area. It became a movie house, then a Broadway theater. The Palace opened as a vaudeville venue in 1913, at a time when the invention of neon lights was turning the area into a nighttime theater district. The theater’s evolution is a tidy encapsulation of the evolution of the city’s entertainment scene, an economic engine that has always drawn visitors to New York. |
